Structural characterization of the RNaseH2 PCNA complex and its implication in replication. We report the crystal structures of a PCNA:RNase H2 complex from Archaeoglobus fulgidus and the human PCNA:RNase H2-peptide complex. The principle interface for both structures is the binding of the PIP-box motif RNase H2 to a hydrophobic pocket of PCNA. The full-length complex illustrates three possible binding modes that position the active-site of the enzyme at different heights above the center of the PCNA ring. Biochemical characterization of the role of PCNA on RNase H2 activity will be also discussed.
