Susceptibility to progressive NAFLD

Susceptibility to progressive NAFLD NAFLD is a paradigm for a complex genetic disease trait; subtle inter-patient genetic variations and environment interact to determine disease phenotype and progression. In recent years, genome wide association studies and candidate gene association studies have identified a wealth of modifier genes that influence different aspects of NAFLD pathogenesis and thus progression. Particular interest has focused on the role of PNPLA3 although the mechanisms of action remain elusive. Knowledge of the genes that determine variation and their biological function enable us to better understand pathogenesis, identify new pharmacological targets and, by adopting a 'stratified medicine’ approach, may soon allow clinicians to identify those at greatest risk of morbidity so that treatment may be directed to those who will benefit most.
